Phenotypic and Molecular Analysis of Mutants Generated with Ion Beam in Tobacco (Nicotiana tabacum)

Abstract
The anthers of tobacco (Nicotiana tabacum cv. Bright Yellow-4) were irradiated with ©ö©ùN-heavy ion beam at 5 Gy, and then haploids were selected in M©û generation (pale-pink flower color). The haploids of M©û generation were self-pollinated and flower color mutants were selected from M©ü generation. The phenotypic and molecular differences were observed between the mutants and the non-mutants. The petals of the mutants were rounder in shape and had much lighter color compared to those of non-mutated ones. The dot blotting and Northern hybridization analysis revealed that the chalcone synthase (CHS) gene in flower tissues was less expressed in the mutants than in the non-mutants. This implies that the expression of CHS gene in the anthocyanin biosynthesis pathway could be suppressed in mutant plants.
